diagnosis of ornithobacterium rhinotracheale using polymerase chain reaction (pcr)
Authors
abstract
abstract: ornithobacterium rhinotracheale (ort) has been identified as one of the emerging respiratory bacterial pathogens in turkey and chicken flocks. diagnosis of ort infection has been faced some difficulties in isolation and identification of the bacterium based on the biochemical properties. a reliable identification method that can be used in routine laboratory investigations is of importance. the purpose of this study was diagnosis of ort using polymerase chain reaction (pcr) in comparison with culture. the pcr was optimized using the primer combination or16s-f1 and or16s-r1, positive control of ort bacteria, and 7 other bacteria such as haemophilus paragallinarum and pasteurella multocida as negative control. all samples were prepared for dna extraction by use of phenol- chloroform method. sixty pooled tracheal and infraorbital sinus samples from 30 broiler flocks slaughtered at an abattoir in gazvin province were examined for presence of ort using culture and pcr assay. a fragment of 784-bp was amplified in 5 positive known ort samples, but not in other 7 bacteria as negative controls. 19 out of 60 primary samples including 11 homogenized tracheal samples and 8 infraorbital sinus swabs, and also all 17 suspected and purified colonies identified microbiologically as ort were positive in pcr assay. one out of 41 negative primary samples in pcr test became positive through the culture. then the propagated bacterium was confirmed in pcr. fifteen out of 30 flocks (50%) were positive in pcr test and only one of them was negative in culture. upon the results the pcr method of this study can be used as a reliable and rapid identification of ort in samples suspected to ort infection. however, it is better to use a combination of the pcr and culture in order to maximize the diagnosis of ort infections. key words: diagnosis ornithobacterium rhinotracheale (ort), polymerase chain reaction (pcr), broiler.

Polymerase Chain Reaction (PCR)
The polymerase chain reaction (PCR) is a powerful and widely used technique that has greatly advanced our ability to analyze genes. Genomic deoxyribonucleic acid (DNA) present in cells contains many thousands of genes. This makes it difficult to isolate and analyze any individual gene. PCR allows specific DNA sequences, usually corresponding to genes or parts of genes, to be copied from genomic...
full textPolymerase Chain Reaction for the Diagnosis of Tuberculosis
Dear Editor-in-Chief We read with interest the study by Khazaei et al. (1) in which the authors have nicely concluded that PCR is more sensitive test than Ziehl-Neelsen staining and histo-pathological examination for the diagnosis of tuberculosis (TB). They have rightly pointed to use PCR, selectively, in acidfast bacilli negative paucibacillary forms of TB. However, we intend to highlight few...
full textDiagnosis of bacteremia in patients with suspected septicemia using polymerase chain reaction method
Background: Sepsis or blood stream infection is a clinical lethal syndrome with severe systemic inflammatory response to infection, if not treated quickly, is associated with dangerous consequences and high morbidity and mortality. The traditional and conventional method for identification of sepsis is blood culture method which is so time-consuming and long that it eliminates the possibility o...
full textVaccination of chickens against Ornithobacterium rhinotracheale infection.
Vaccination of young broilers with inactivated vaccines against experimental Ornithobacterium rhinotracheale challenge was found to be effective, but the results of vaccination were influenced, in a negative way, by the presence of maternal antibodies. The use of a strong adjuvant, such as mineral oil, in a bacterin was necessary to obtain good protection when maternal antibodies were present. ...
full textDevelopment Application of Polymerase Chain Reaction (PCR)
Polymerase chain reaction (PCR) is an in vitro technique to synthesize large quantities of a given DNA molecule that separates the DNA into two complementary strands, uses a peltier heat pump to quickly heat and cool the DNA and uses the Taq polymerase for the synthesis of DNA. Taq is a bacterium that lives by volcanic sulfer jets at the bottom of the ocean where the temperature is very high. F...
